摘要(中) 本研究利用表面增強拉曼散射(Surface-Enhanced Raman Scattering, SERS)分析纖維化的肝細胞,希望能以這種光譜技術量化肝細胞纖維化的程度。由於物理拉伸的方式無法進行定量分析,我們在肝細胞樣品中添加不同濃度的化學藥劑(Transforming growth factor beta, TGF-β),以得到不同程度的纖維化細胞。根據文獻,膠原蛋白的過度沉積是肝纖維化的一個主要特徵,且1315 cm-1為膠原蛋白拉曼共振所造成的特徵峰。在實驗中,我們觀察到TGF-β添加下肝纖維化細胞在相關特徵峰強度上呈現成長趨勢。此外,為了進一步深入了解纖維化的程度,我們增加了放置時間,結果顯示隨著時間的延長,纖維化的程度越明顯。SERS是一種高速、精準、非侵入性的檢測技術,本研究以SERS確認了TGF-β濃度、TGF-β浸泡時間和肝細胞纖維化之間的關聯性,有潛力應用至臨床診斷。
摘要(英) This study employs surface-enhanced Raman scattering (SERS) to analyze the fibrosis of human hepatic cells. The goal is to investigate the impact of chemical additive (Transforming growth factor beta, TGF-β) concentration on the behavior of liver fibrosis cells. Since the physical means (stretching) was not suitable for quantification analysis, we immerse the cells in the solution with varied TGF-β concentrations, aiming to see the degree of cell fibrosis vis the SERS spectra. According to the literatures, excessive deposition of collagen protein is a key characteristic of liver fibrosis, with the Raman characteristic peak at 1315 cm-1. In the experiment, we observed the increased SERS intensity at 1315 cm-1 with the liver fibrosis cells in TGF-β. Furthermore, to gain a deeper understanding of the extent of fibrosis, we extended the incubation time, and saw that the cell fibrosis becomes more pronounced with increased incubation time. These findings provide important insights into the relationship between TGF-β concentration, incubation time, and cellular responses in liver fibrosis.The SERS technique, with the advantages of high throughput, high sensitivity and non-invasion,is of high potential for clinical applications.
