;The sable1 is a classic X-linked hypomorphic mutation whose causal locus was originally unspecified. To identify the causal locus, a series of chromosomal deletion strains was generated by FLP-FRT recombination technique for complementation test, and performed RNAi-mediated knockdown of the candidate genes. The further verification results by Sanger sequencing data for gene mapping suggesting that the sable1 mutation resides within the Yippee gene locus. Motivated by a fundamental interest in the poorly understood functions of Yippee and preliminary observations of physiological impairments in our laboratory strains. A series of systematic evaluation was conducted of its impact on adult climbing ability, longevity, and female fertility. By introgressing the Ypes-1 allele into three distinct genetic backgrounds—153.6, Oregon-R, and w1118—Cuticular hyper-melanization and a prominent dark thoracic trident pattern were consistently induced by Ypes-1, regardless of the genetic backgrounds. Furthermore, the mutation caused a profound reduction in longevity and female fertility; the latter associated with stunted egg chamber development and reduced ovary size. Notably, while climbing ability displacement diminished in 153.6 and Oregon-R Ypes-1 mutants, this effect was reversed in the w1118 background, where w1118, Ypes-1 double mutants exhibited significantly stronger climbing ability than w1118 controls. In summary, these findings provide a more comprehensive profile of the pleiotropic effects associated with Ypes-1, establishing that Yippee is critical for locomotion, aging, and oogenesis, in addition to its established role in cuticular pigmentation.
