;This study develops a multifunctional composite nanoparticle, γ-Fe₂O₃@SiO₂@Au, which exhibits both superparamagnetic properties and localized surface plasmon resonance (LSPR)-induced photothermal effects, serving as a nanomaterial platform for photothermal-driven nucleic acid amplification. For material synthesis, superparamagnetic iron oxide cores (γ-Fe₂O₃) were first prepared via thermal decomposition. To enhance colloidal stability in polar solvents and provide surface modification sites, a thickness-controllable silica intermediate layer was coated onto the cores using the inverse microemulsion method. Subsequently, the silica surface was aminated, and negatively charged gold seeds were uniformly grafted onto the magnetic beads via a click chemistry reaction between trans-cyclooctene (TCO) and Tetrazine (Tz). A uniform gold shell was then successfully constructed around the nanoparticles using the seed-mediated growth method. Furthermore, the magnetic core enables rapid magnetic separation and enrichment, which facilitates subsequent biological sample pretreatment and analytical integration. Finally, a laser-driven photothermal testing platform was established, verifying that the composite nanoparticles possess excellent temperature control capabilities for both stable isothermal maintenance and repeatable thermal cycling. In conclusion, the magnetic surface plasmon nanocomposite and the photothermal platform established in this study lay a solid foundation for photothermal nucleic acid amplification, offering great potential for future applications in thermal cycling and isothermal nucleic acid detection.
