;To address the demands for high sensitivity, multiplex recognition, and signal encoding in biochemical analysis and cancer biomarker detection, this study developed a surface-enhanced Raman scattering (SERS) encoded nanoprobe based on silver mesocrystals (AgMCs) for multiplex detection of cancer-related biomarkers. AgMCs consist of ordered assemblies of nanocrystals, combining the multiscale surface features of mesocrystalline structures with the localized electromagnetic enhancement properties of silver, making them suitable as SERS substrates for encoded nanoprobes. In this study, AgMCs with distinct surface structural characteristics were prepared through morphological control. Their surface plasmonic properties and the distinct spectral signatures of Raman reporter molecules were exploited to construct encoded probes. By incorporating different Raman reporter molecules, a library of seven spectrally encoded SERS probes was established. These probes were identified and analyzed based on the characteristic Raman peaks of the respective reporter molecules. The results demonstrated that the encoded probes generated distinct Raman spectral signatures, enabling their application in multiplex SERS analysis. For biomarker detection, the encoded SERS nanoprobes were applied to the detection of prostate cancer-related biomarkers, with target recognition achieved through the characteristic signals of different Raman reporter molecules. The results demonstrated the feasibility of using the probe system for SERS-based analysis of cancer-related biomarkers, with potential for multiplex detection and spectral encoding. Overall, this study established an AgMC-based encoded nanoprobe platform and demonstrated its application in the multiplex detection of cancer-related biomarkers in vitro.
