;Cells are regulated by various physical stimuli in vivo, including mechanical forces, electrical signals, and fluid flow, all of which influence cellular growth, differentiation, and function. However, conventional static culture systems are unable to accurately reproduce the complex physiological microenvironment of living tissues. Therefore, developing advanced cell culture platforms with enhanced physiological relevance has become an important research objective.
In this study, a previously developed modular multi-physical stimulation bioreactor was further optimized to improve system stability, operational flexibility, and stimulation parameter control. Mechanical stretching, electrical stimulation, and automated scheduling functions were integrated into a Raspberry Pi-based control system with a graphical user interface, enabling user-friendly operation and improved experimental reproducibility.
Based on the optimized bioreactor, microfluidic and perfusion culture concepts were introduced to miniaturize the platform into a Muscle-on-a-Chip system. The chip was fabricated using polydimethylsiloxane (PDMS) and incorporated a conductive polypyrrole (PPy) layer for electrical stimulation. The resulting platform provides mechanical stretching, electrical stimulation, and continuous medium perfusion within a compact and integrated system while reducing device size and culture medium consumption.
