    Abstract: 基因調節的變異已成為許多重要的研究主題方向,如癌症、演化、種化、...等等;若要拿來做為臨床上的應用,以治本的目的而言,最好要能瞭解其完整的調節機制。在過去由於傳統研究技術的限制,其進展速度很慢;但現在可以藉由最先端的技術,如Pyrosequencing或Solexa,並運用在已有較充分研究的模式生物,如酵母菌(Saccharomyces cerevisiae),如此的配合即可提高對未知領域(如基因調節的網路)研究的效率及精確性,因此本項計畫便利用這些優點針對內因子的變異進行探討,首先找出關鍵的核甘酸位子,再找出結合在此核甘酸的轉譯子,如此即可解讀出內因子的基因調控機制;同時,這些因子的變異性可進而幫助我們深入探討其在基因演化上的意義及重要性。 Gene regulation becomes a central issue to many major subjects, such as cancer, evolution, speciation … etc. To apply this knowledge on clinic heavily relays on the understanding of the detail mechanisms. Due to the limitation of traditional technologies the progress is very slow. Now, with the most advance modern technologies, like Pyrosequencing or Solexa, plus a well studied model organism, as yeast (Saccharomyces cerevisiae), these make the gene regulation studies much more efficient and accurate. Therefore, this study taking the advantages of these technologies and knowledge to focuses on cis element variants in order to reveal some key nucleotides for the gene regulation mechanisms of detected genes. With those key nucleotides we can further detect which transcription factors (TF) binding on them. Consequently, we reveal the mechanisms of gene regulations at their cis region. Also, these cis variations represent substitution information which may assist us to have insight into their evolutionary significance. 研究期間 : 9808 ~ 9907
