    Abstract: 本計劃係有一種光纖式條碼載體及微流道晶片式流式細胞儀生物體檢測裝置,其主要係將生物粒子(如:蛋白質、抗體、細菌或去氧核醣核酸)加於光纖載體(carrier)上,該光纖載體設有條碼,並將此含生物粒子之光纖載體注入本計劃晶片式微流道細胞儀 (micro-flow cytometer ,MFCM)中,利用水壓力或吸力將此載體流至為通道細胞檢測,裝置中偵測器前並由該偵測器經我們所設計隻電腦影像識別程式讀取載體上隻條碼 (barcode),已得之微通道流式細胞儀所分析之生物粒子的類別、數目及其他生物參數。生物晶片式微流道細胞儀乃是利用LIGA-like MEMS 技術,以準分子雷射(EXCIMER LASER)經光罩圖案製成。 This proposal provides a method for producing a Novel type fiber coded micro-carrier and micro flow cytometer chip detection system. A simple and cost effective solution for bio-molecule applications was developed. Application relevant items such as manufacture process, biospecific interaction, and analysis method are discussed. For low cost fabrication, the use of LIGA-like process is suggested. LIGA-like process is used as a dry mask patterning process in which an intense beam of light from an excimer laser through is to pattern material directly. This process has found extensive application in the microelectronics industry for patterning of polymer materials. The use of LIGA-like techniques also offers two another attractive features: first, we can cut the fiber polymer into many tiny micro- fiber carriers with micrometer precision. Second, LIGA-like process allows to encode with high precision spatial information onto the micro- fiber carrier that can be used in the identification of the bio-molecule. This proposal gives a description of the good idea, describes the fabrication of the novel micro- fiber carrier that we called 「coded micro- fiber carrier and active biochip micro flow cytometer,」 and of the image processing algorithms used for the analysis of bio-molecules. This study also provides a test method for identifying a bio-molecule, which includes mixing several coded micro-carriers with the hybridized unknown bio-molecules; and identifying the codes on the micro-carrier via image recognition system. The numbers and types of the known micro-carrier can be flexibly adjusted according to the number of tested bio-molecules. 研究期間:9308 ~ 9407
